Sea World is definitely one of the Orlando area parks that you should try and get to see. They have spectacular shows and is an overall great park.
Now if you want to get there by public transportation you can, but the best way I believe if you are staying at a Disney resort is to take the free Disney transportation to Downtown Disney then on the street beside there you can get on the city bus system (not free) and get to the park. Most of the buses I saw with the city system was wheelchair accessible, I only came across one bus that was not wheelchair accessible. All the Disney transportation system is wheelchair accessible.
Now some of the city bus routes do not operate late into the evening, we where only there in the daytime and did not stay into the evening so we did not have an issue, but if you intend on staying to closing (on a day they are open late into the evening), make certain that you check http://www.golynx.com/ and see the routes necessary to get there and back and the schedules prior to visiting sea world. If there is an issue with transportation, you can always be prepared to take a taxi back (the more costly option compared to public transportation) if you do not have your own vehicle.
Wheelchair accessibility at the park is no problem except for some of the amusement rides which are not wheelchair accessible.
